I am growing human primary B cells in the CD 40 system and currently have
two technical problems:-
1.Replicates in 3H thymidine uptake assays are very poor. I think this may
be due, at least in part, to the cells growing in clumps. I don't have a
problem with my standard assays, just in the CD 40 system.
2.After differentiation with IL-10, we see levels of IgG secretion which
are comparable to the published data but I am not detecting any significant
levels of IgM. The Elisa assay used for detecting IgM is working O.K. Any ideas?
