polyclonal antibodies in mice

Our lab has made hundreds of poly clonal antibodies in mice, mostly in 
the past so dim only I remember it.  The only reason to use a mouse is to 
make a hybridoma after ab is porduced.  Some one with a lot of proactice 
can bleed a mouse every other day for 4 times rest a repeat it for 
months, it is not a good way to make the antibody.  At best you can get 
2-300ul of blood from a mice yielding 100ul of serum/bleed.  Change your 
detection kits.  Its cheaper in the long run.  If you don't do this get a 
company to make the antibody for you.  It saves filling out all the 
animal forms.  When we did this we immuniozed froups of 10-15 mice, and 
pooled serum.

To directly answer your questions 10-100ug/mouse of protein in adjuvant 
(we have had pretty good luck with RIBI)  three or four times two to 
three weeks apart its usually pretty effective for inducing antibody.
BTWcost for a BALB/c from Jackson is $18.00/mouse.  Can this really be 
cost effective.
