Thanks for the RACE reference, but you'll note that the procedure I
described is a significant modification of the original procedure
(reduced background due to use of an amine-modified adaptor which is
used both for 5' and 3' RACE, and combination of the 5' and 3' RACE
reactions to get the entire cDNA, using LD PCR).
I've spent months trying to get 5' ends of some tough cDNAs, so if
this procedure works I'll use it. But I refuse to overpay for a kit.
I'll figure out the nitty gritty myself, but a literature reference
would sure help.
