I Mikael Boesen, have during the past 2 month tried to produce anti-lyt2
antibodies as ascites in NMRI nude mice. My first problem has been, that the
cell lineage (rat anti mice hybridoma) from ATCC, cath. no 53-6.72, was
making solid tumors. In order to overcome this problem I have, with succes,
converted the cell lineage to ascitesproduction by teasing cells into
suspension and reincerted them into another mice.
Questions!!
1) How do I cryopreserve the ascites, so that I can use it later in in vivo
experiments?
2) Does pristanetreatment of the mice before cell injection, have any effect
upon the antibody concentration in the ascites.
3) Is there another way, beside ascites and spinner, to produce large
quantities of antibody.
If you are able to answer any of these questions, I would be very greatfull
if you could give me the references or protocols where I can look it up.
