I would appreciate some good advice on the following problem:
I need tons of Monocytes (at least 10e8) from human buffy coats. What
method is useful and comfortable handling 5 or more buffy coats for
isolating as many monocytes as possible (purity at least 90 %). In a first
experiment I tried to isolate them from Ficoll-purified PBLs using the
MACS-system (CD14). Unfortunately most of the monocytes rushed through the
column and the yield was no more than 6e6 cells from 5 buffies.
So how do you out there isolate your monocytes ?
