You do not have any help (ie CD4 stimulation). Spleen cells work because
dentritics etc. stimulate CD4 which help CD8 via cytokines primarily.
You may try adding allo CD4 stimulators (and later sort) or try adding
ConA stimulated sup to your hep/CTL culture.
