The best method (in my experience) for separation of PMNs from human blood is
lysis of RBC in isotonic NH4Cl followed by differential centrifugation,
described in :Eggleton et al, J.Immunol.Meth 121, 1989, 105-113
It gives much higher yield than using separating media, which could be
important with small volumes of blood.
BTW, it has to be fresh blood, not serum.
