Hi Danny,
There are some informations that you need to include in the question,
especially what construct you have been using for display; is it a phage
or a phagemid vector?
If the latter is the case, you will probably have monovalent display, i.e.
one displayed scFv per phage particle. If so, the number can be increased
to 3-5 copies per phage by switching to a phage system which will result
in display on all pIII on the phage.
If you are using a phage vector already, pVIII COULD be a solution, but
this would hve to be in a phagemid vector so that wild type pVIII is
present. I cant say if it is possible though... Maybe somebody else
knows of the size limitations for pVIII fusion? Peptides hve been displayed,
but 27 kDa is another matter.
