I have several versions of Excel spreadsheets set up to calculate LDA by
Chi Square minimization methods. The line fitting is done by a macro.
You can cutomize the number of replicate wells and the cell input numbers
easily. Changing the number of responder cell concentrations plated may
take a bit more work but can be done. One version is used to calculate
total clonable T cell LDA's we do to monitor TCD bone marrows. The number
of negative wells is entered into the spreadsheet along with cell counts
before and after treatment and flow data on the predepleted same in order
to correct for cloning efficiency. The data is also graphed (with 95%
confidence intervals) and a p=value for the line fit is calculated. Two
others are designed to accept data input one from a gamma counter, and the
other from an ELISA reader to do CTL and HTL LDA respectively. If you are
interested in any of these 3 spreadsheets let me know and I will send them
to you via e-mail attachment. They will run on either a Mac or a PC.
