I have joined this thread late, so sorry if I have missed something.
As far as I know, while Con A is a metalloprotein (needing bound Mg/Mn)
as long as these are not stripped from it (with high concns of EDTA, low
pH treatment, whatever) it will bind to target proteins fine in the
absence of exogeneous Mg/Mn.
