Im looking for some advice concerning the best way to do intracellular
cytokine/surface marker staining (three colour). Basically I have tried it a
few times using FACS perm (BD) to permeabilise the cells (antigen stimulated
human PBMC) and anti-cytokine/CD marker antibodies (BD) with reasonable
results. We are planning to expand these experiments and as a result I need
to buy in the antibodies and check the protocol for this type of staining.
Although these experiments have been relatively succesful so far I want to
cover all options before paying out £3000 for all the reagents we will need
to use.
The alternatives I have in mind are Dako and Coulter and I am planning to
get in touch with these companies to see what the have on offer. However any
advice would be appreciated.
