We routinely use MACS (by Miltenyi Biotech) for T cell isolation by negative
selection and get >95% CD3-positive cells.
What are you sorting and for what purpose? MACS is probably better for
handling large numbers of cells in a relatively short period, but sorting by
cytometer will probably give you better purity. Miltenyi also markets MACS
as a "pre-sort" isolation step prior to sorting by cytometer for small
populations (ex. CD34).
