TCR transgenic

Hi,
I was wondering if anyone could give me any advice on some TCR transgenic
mice we had generated. These mice are transgenic for a Va2, Vb4 TCR that
recognizes a 9-mer peptide (77.2) derived from a T. cruzi protein called
TSA-1 in the context of H-2Kb. We have Kb-77.2 tetramers which bind a
fairly large percentage of our lymphocyte gate (in both peripheral blood and
spleen), however very few of these cells express CD8 or CD4, and most are
double negative. So far I have been unable to see any peptide specific
function, either. Does anyone have any suggestions for backcrossing or any
theories about why these cells are not selecting properly?

Thank you,
Jennifer Kraft
