I knew it is unclear that antigen retrieval methods with high temperature
is to break the bonds that were formed during formalin fixation. But
lately I have tried CD3 antigen retrieve Microwave Oven in pH9.0 buffer
(10mM tris.HCl, 1mM EDTA), what is the mechanism? I get good staining,
especially in small intestine, also in inflamed pancreatic islets. But I
may need to see more T-cells in inflamed islets. I need to optimise the
condition, it may help if I knew more the mechanism of the antigen retrieve
in pH 9.0.
