We are trying to establish two stable cell lines using G418 resistance and hygromycin resistance. After determining the concentration of each drug needed to kill the cells, we proceeded with transfections. Some of the control cells (approx. 30%), those transfected with no DNA, did not die. Has anyone encountered this problem before? If so, what actions did you take? Could you still establish a stable cell line?
