I'm not quite sure what you are trying to do, but must you digest the
scaffold prior to performing ELISA? Are you concerned about getting
access to ECM trapped within the scaffold? Otherwise, I would think you
should be able to perform the ELISA directly on the scaffold itself.
Also, most ELISA's use antibodies that recognize peptides (fragments)
so often even denatured proteins can be detected (this varies from Ab
to Ab, however, so you should check with the ELISA manufacturer if
using a kit or Ab supplier if making your own). Also, if I'm not
mistaken, I had thought that PLLA was suspectible to hydrolysis, which
would make me think even a mild aqueous environment should do the job
without bothering your ECM proteins much. Hope that helps.
