I used to run polysomes  ages ago, including some from with HeLa cells
(see: Mol Cell Biol. 1987 December; 7(12): 4357-4368, for HeLa; also J
Biol Chem. 1999 Apr 23;274(17):12081-6 for other cells). It's going to
hard to tell what's wrong until you give us some details on what you
are doing.
