For my masters project I am looking at the protective roles of a number of
compounds on neurons affected by glutamate toxicity.
I wish to micro-inject glutamate, NMDA, or Quinolinic Acid into the brain
and a few days later examine the brain and see if there are any histological
differences between the contol brains and those pretreated with my
compounds. I have seen a paper where this has been done before and the
hippocampus was examined.
My problem is that I do not know what concentrations of these toxic agents
to use. I have tried 2ul of Quinolinic Acid (30mM) into the right
ventricle, but I do not seem to be getting any conclusive results.
If anyone can help by giving ideas of what concentrations to use, what
agents would be good to provide maximal neuronal damage, and the areas of
the brain to inject these into, I would be most grateful.
