Your questions about yeast and toxicicty tests were a little vague. What
type of tests? What types of agents?
There are a number of yeast mutants available from the Yeast Genetic Stock
Center at the University of California at Berkely (e-mail:
ygsc305 at violet.berkely.edu). These are available for a very modest fee (~
$10/strain). You might also search the American Type Culture Collection
(http://www.atcc.org).
Workers in the yeast field have described a number of mutations that affect
intracellular levels of metals, steroids, and other compounds. There is
considerable data on pleiotropic or multiple drug resisitance genes that
mediate xenobiotic export in yeast--see the work of Elizabetta Balzi, Andre
Gaffeau, Kieth Yamamoto, and others (review, Biochemica et Biopysica Acta
1073: 241-52, 1991). There are also chitin (cell wall defect) mutants
available, which may be more permeable to some agents.
In studies with exposures of TCDD, benzo(a)pyrene, and other very
hydrophobic agents, I have found that using glass rather than plastic
culture tubes greatly improves getting these compounds into yeast. I
presume that the hydrophobic compounds are interacting with the plastic
surface rather than remaining in solution. Note that yeast media, in
comparison to mammalian tissue culture media, is very different in
composition (e.g., protein content) and is of lower pH.
