Hello netters,
Please pardon the rather simple nature of the post but I hope to
save some time by learning from other people's experiences...
Basically, what is your favourite/easiest/cheapest/most effective way of
blunting a DNA fragment? - with 5' and/or 3' overhangs
At the moment I use a protocol from Basic Methods in Molecular Biology
that involves incubating the sample with Klenow, first in the absence and
then in the presence of nucleotides. I realise that Klenow will work
for both 5' and 3' overhangs but is most efficient with the former.
Can you mix Klenow and T4 if the DNA has one 5' and one 3' overhang?
Can you recommend a better protocol? Have you encountered any problems
with the one I have outlined - have T4 or Klenow failed with a particular
overhang?
Generally the protocol works fine for me but I have run into problems
in the past and was wondering if there was anything better out there.
