I would like to know if everybody who got these blue glowing gels used the
same TAE-recipe as John
: 50X
:
: Trizma HCL 228.8 g
: Trizma base 66.4 g
: Na acetate (anhydrous) 82.0 g
: 0.5 M EDTA, pH 8.0 200 ml
: dH2O to 1 litre.
I prepare may 50xTAE as described in Maniatis, Tris base, acetate, EDTA
(no Na+, no Cl-) and didn't ever see this blue glow though sometimes using
rather old 1xTAE, running it for several times, sometimes running it very
hot and preparing the gels by boiling in the microwave.
May be this slight difference in buffer composition leads to problems under
certain conditions.
Cheers
  Klaus
