Oligo Madness

Hi Folks,

I'm designing oligos for mutagenesis. The mutagenesis will be done by the
polymerase method (eg Stratagene 'QuikChange'). It is, apparently,
advisable to have C or G at each end of the oligo.

The end of one of my - putative - oligos looks like ...CCATCACCC. I know
with sequencing that G/C clamps at the end are a Bad Idea. What worries
me is that 3Cs at the end might cause mis-priming. Would ...CCATCAC
work better? The latter case has 9 parent bases after the last mutant
site, which is the bare minimum for effective hybridization. Does anyone
have any comments?

TIA,

Richard
