Keven,
What is your template isolation method. If it is Quiagen or
alike with a solid support column, I have seen where some of this
support can come through into samples. These then pellet down with
ppt DNA and will not resuspend when dissolving your plasmid. If you
are using these methods of template prep then try a control of just
H2O over the column and see if you get solid pellet/ppt. Hope this
