Regarding Plasmid Expression

Dear all,I have been working with a gene cloned into pcDNA 3.1 V5/His
vector. However, over time the expression has decreased drastically.
However, another gene cloned into same vector has consistently high
expression, till now (that rules out cell problems too). I did prepare fresh
Midi stock..but problem remains. What has gone wrong?
