Problems with P1 transduction

Hi Folks,

We are trying to move the delta(lac)U169 proC::Tn5 mutations from
Y1088 into a rec+lac+pro+ E.coli using P1Tn9clr100. The recipent
strain is OK in that other transductions into it work fine but we get
nothing at all from lysates prepared on Y1088. Our Y1088 is kan
resistant and lac- so that's OK. We have noticed that Y1088 takes
much longer to lyse (4 hours) than other E.coli's we have used. Maybe
the titre is still too low but we have tried six separate lysates now
with no success. Could the supF of Y1088 be screwing it up? Any
ideas or do you know of another rec+ proC::Tn5 lac- strain we can
try?

Finally what host strain does one use to titre the lysogenic P1Tn9clr100
phage.

Many thanks

Duncan
