U9 MEDIUM   FORMULA FOR 1 LITER

TRYPTIC DIGEST BROTH         7.5 gm
SODIUM  CHLORIDE                      5 gm
KH2PO4                                        0.2 gm
HORSE SERUM                            50 ml
UREA SOL'N 10%                           5 ml
PHENOL RED SOL'N 1%               1 ml
PENICILLIN G 100,000u/ml          10 ml
AMPHOTERICIN B                     250 mg

FOR U9-B MEDIUM    ADD  5ml 2% L-CYSTEINE SOL'N

FINAL pH = 5.5

My suggestion....prepare it and filter sterilize ...Method #2
sterilize base (components 1,2,3 &6) add rest asepticlly.
