I am not aware of AO being affected by methanol fixation although I
usually use a Hotchkiss McManus Schiff's stain for image analysis to
dtermine ploidy (which I assume you are doing?). You might like to use it
to cross check your results although I assume you are already using a
control. We routinely used 50% methanol for fixing our urine cytology
specimens for DNA image analysis
