RESOLVING MICROSATELLITES ON METAPHOR AGAROSE
Has anyone out there tried resolving microsatellite alleles on Metaphor
agarose rather than the standard polyacrylamide? Metaphor is claimed
to resolve differences of 1-2% of the fragment size. Since my
microsatellites are around 100bp, they need to run quite far and hence
get diffuse and faint. What are the tricks to resolving on Metaphor, or
am I wasting my time? Do I need more PCR product, narrower-toothed combs or
what?
I would appreciate any feedback
