Measuring Intracellular Calcium Concentration

I am attempting to evaluate the effects of manipulating 
the intracellular calcium concentration of VMA-6 (vacuolar 
membrane ATPase) mutant yeast cells.

Currently, I have used FURA-2AM to measure the [Ca2+].  Now I 
am trying to use FURA-2 and DMSO (dimethyl sulfoxide).  Where 
can I find information on using these chemicals?  Is there a 
better method of measuring the Calcium concentration?
