I'm not sure about vaccinia, but I do RT/PCR analysis of influenza virus from plaques all the time. Just blow apart the virus (I do a SDS/PK and phenol/chloroform extraction) and throw in the goodies. Neil. Neil Parkin nparkin at aol.com
