Dear buddies,
There are many ways how to make yeast mRNA and everyone considers his/her
the best one. Our whole lab is now since 2 years happily using the
technique published by Cross and Tinkelenberg, Cell 65:875-883, 1991,
which is a very fast and reliable method based on mechanical breaking
with glass beads directly in the presence of phenol. You just have
to be really careful not to contaminate the post-phenol supernatants
with bits from the interphase (don't be greedy!), the rest is practically
foolproof and considerably less nasty to do than any hot phenol prep.
