Plasmid Isolation from Yeast

i think the key is to electroporate whatever you get out of the yeast into
e.coli to amplify your plasmid.  i tried for a long time (unsuccessfully)
using standard calcium chloride transformations of E. coli.  the yeast
preps are are contaminated with large amounts of genomic DNA which makes
your transformations less efficient.  electroporation provides a much
higher transformation efficiency.  i've had no problems with yeast
plasmids since switching.
