Immunolocalisation in yeast

Hi netters,
I'm doing immunolocalisation on slide (fixation with polylysine) The
background noise is too high. I think it is due to the washing that is
not so powerful on slide. Does somebody have a protocol where the
treatment with antibodies is in tube and not on slide. In other words,
can I do the same in tubes...
